摘要
采用乙酸-胃蛋白酶结合法,从猪腱中提取I型胶原。运用原子力显微镜(AFM)观察了所得I型胶原的形貌特征,用聚丙烯酰胺凝胶电泳法(SDS-PAGE)分析所得I型胶原分子质量的分布。结合傅里叶变换红外光谱(FI-IR)、超灵敏差示扫描量热仪(US-DSC)和圆二色谱(CD)的分析,与市售Sigma I型胶原对比,证明了所制备产物为I型胶原,且很好地保持了I型胶原所具有的天然三股螺旋结构。
Collagen type Ⅰ was extracted from pig tendon with the method of acid swelling- pepsin digestion. Atomic Force Microscope( AFM) was used to characterize the appearance characters of collagen type Ⅰ. The molecular weight of the collagen was determined by SDS- PAGE. In addition,the collagen were measured by Fourier Transform Infrared( FT- IR),Ultra- sensitive Differential Scanning Calorimeter( US- DSC) and Circular Dichroism( CD). And compared with the result of Sigma Ⅰ. Results indicate that the product is collagen type Ⅰ and it sustains original triple helix structure of collagen type Ⅰ very well.
