摘要
目的研究膜型基质金属蛋白酶-1(MT1-MMP)表达对肝细胞癌血管生成拟态(VM)形成的影响及机制。方法构建靶向MT1-MMP的RNA干扰质粒载体,转染肝癌细胞系Bel7402,实时定量PCR及Western blot检测干扰效果;在体外三维细胞培养体系中观察其对VM管状结构形成的影响;Transell侵袭小室实验检测细胞侵袭能力。结果靶向抑制MT1-MMP的表达下调肿瘤细胞侵袭能力,影响肝癌细胞在三维细胞培养体系中VM管状结构的形成。结论 MT1-MMP表达影响肝细胞癌VM的形成,可能成为肝细胞癌抗血管生成治疗的新靶点。
Objective To investigate the effect of MT1-MMP on vasculogenic mimicry(VM) formation in hepatocellular carcinoma(HCC) and discuss its possible mechanisms. Methods RNA interfere plasmid targeting MT1-MMP was constructed and transfected into a HCC cell lines. Interference effect was detected by real time PCR and Western blot. VM networks in 3D cell culture system were observed with a microscope. Cell invasion assay was performed using 24-well transwell. Results Inhibition of MT1-MMP expression impaired VM network formation in vitro by down-regulating cell invasion. Conclusion MT1-MMP influences vasculogenic mimicry formation in HCC cell line and offers a potential molecular target for HCC antiangiogenesis therapy.
