摘要
【目的】优化大白花杜鹃总黄酮的提取工艺条件,为其药用开发提供技术支持。【方法】以大白花杜鹃为原料、乙醇为提取溶剂,通过单因素试验及正交试验考察料液比、提取温度、乙醇体积分数和提取时间对大白花杜鹃总黄酮提取效果的影响。【结果】影响大白花杜鹃总黄酮提取效果的因素顺序为:提取温度>乙醇体积分数>料液比>提取时间,其最佳提取工艺条件为:以60%乙醇为提取溶剂,在料液比1∶60、90℃条件下回流提取2.5 h,大白花杜鹃总黄酮提取率为25.9%。【结论】乙醇回流法具有操作简便、提取率高、稳定性好等特点,可有效提取大白花杜鹃总黄酮。
【Objective 】Extraction technology for total flavonoids from Rhododendron decorum Franch was optimized to provide technical support for its medical development. 【Method 】With Rhododendron decorum Franch as raw material and ethanol as extraction solvent, the effects of solid-liquid ratio,extraction time,ethanol volume percentage and extraction temperature on total flavonoids were evaluated using orthogonal test method. 【Result 】The descending order of influence was determined as following: temperature, ethanol volume percentage, solid-liquid ratio and extraction time. The content of total flavonoids in flower of Rhododendron decorum Franch was 25.9% under the conditions of temperature of 90 ℃,solid-liquid ratio of 1 ∶60, extraction time of 2.5 h,and ethanol volume percentage of 60%.【Conclusion 】Ethanol reflux method is simple and stable with high extraction rate of total flavonoids, hence it is effective for extracting total flavonoids from Rhododendron decorum Franch.
