固本止咳中药对COPD小鼠肺组织γδT细胞及IL-17的影响 被引量:15

摘要 目的:观察固本止咳中药对COPD模型小鼠肺组织上皮细胞间γδT细胞及IL-17影响,探讨其治疗COPD的作用机制。方法:采用鼻腔滴入LPS加熏香烟的方法建立COPD小鼠模型,实验分为空白对照组、模型对照组、固本止咳中药组;运用HE染色对形态学进行观察,评价COPD小鼠模型;应用流式细胞术检测肺组织上皮间γδT淋巴细胞表达情况;并应用ELISA方法检测小鼠肺组织上皮细胞间淋巴细胞培养上清液IL-17分泌水平。结果:COPD小鼠模型形态学结果符合COPD临床病理形态学改变;模型对照组较空白对照组γδT淋巴细胞比例明显升高(P<0.05);中药治疗组较模型对照组γδT淋巴细胞比例明显降低(P<0.05)。淋巴细胞培养上清液中IL-17含量与γδT细胞变化趋势同步。结论:固本止咳中药通过调节γδT细胞的生物活性,减少IL-17的分泌,从而减轻气道炎症,减少气道壁损伤-修复过程的发生,改善了呼吸道黏膜免疫功能。 Objective: To observe the effects and mechanism of Gubenzhike recipe on epithelial gamma delta-T cell in lung and IL-17 of COPD mice. Methods: The mouse COPD model was established by exposure to cigarette smoke in combination with intra- nostrils injection of lipopolysaccharide. The mice were divided into three groups: negative control group, model group and Gubenzhike recipe group; Use the methods of HE eosin staining to observe the morphology characteristic of COPD model; Use the method of Flow cytometry to test the expression of epithelial γδ T cell in lung. Use the ELISA to test the IL-17 level in supernatant of epithelial lymphocyte culture, Results: The morphology characteristic was Up to COPD; The proportion of γδT cells of model group was highel: than negative control group (P〈0.05); And the proportion of γδT cells of Gubenzhike recipe group was lower (P〈0.05); The IL-17 level in supernatant of epithelial lymphocyte culture coincided with the changing trend of γδT cell.Conclusion: Gubenzhike recipe might improve respiratory mucosal immune function by regulating the biological activity of γδT cells, which reduce the secretion of IL-17, relieve airway inflammation and reduce damage and repair process of the airway.
