摘要
目的:检测溃疡性结肠炎大鼠肺肠组织TGF-β1/Smads信号蛋白含量,探讨其肺损害的机制。方法:SD大鼠60只随机分为正常组(30只)和模型组(30只),模型组构建三硝基苯磺酸诱导的溃疡性结肠炎动物模型,正常组大鼠予等量0.9%氯化钠溶液灌肠。实验过程中观察大鼠一般情况,首次造模后第8、29、50天取肺、结肠组织行HE染色观察病理形态改变,电镜观察组织超微结构改变,免疫组化法检测肺肠组织TGF-β1/Smads信号蛋白表达。结果:与正常组比较,模型组大鼠肺、肠组织均出现病理形态改变;模型组大鼠第8、29天结肠组织TGF-β1、Smad3表达含量显著升高(P<0.01,P<0.05),第29、50天肺组织TGF-β1、Smad3表达显著升高(P<0.01,P<0.05)。结论:溃疡性结肠炎模型大鼠出现肺部损伤,其损伤程度随结肠病变进展而加重,TGF-β1/Smads信号蛋白可能通过对结肠和肺的损伤修复作用介导"肠病及肺"的病理传变过程。
Objective: To observe the content of TGF- β1/Smads protein in lung and colon tissues of ulcerative colitis rats and to explore its mechanism of lung injury. Methods: Sixty SD rats were randomly divided into control and model groups, 30 rats in each group. Rats in model group were used to establish the ulcerative colitis model induced by trinitro-benzene-sulfonic acid. The rats in control group received gavage administration with the same amount of 0.9% sodium chloride solution. The general condition of rats was observed in the modeling process. The pathological morphological changes of lung and colon tissues of rats on the 8th, 29th, and 50th day were observed by HE stain and electron microscopy was used to test the expression of TGF-β1/Smad signaling proteins. Results: Compared with control group, the pathological morphological changes of lung and colon tissues of rats in model group were significant. The expression of TGF- β 1 and Smad3 in colon tissues tissue of rats on the 8th and 29th day was increased significantly (P〈0.01, P〈0.05), and the expression of TGF- β 1 and Smad3 in lung tissues tissue of rats on the 29th and 50th day was increased significantly (P〈0.01, P〈0.05). Conclusion: The lung of model rats is injured, and the degree of injury is aggravated with the development of colon disease. The signal protein of TGF- β1/Smad3 could mediate the pathological change process of ‘bowel disease involved lung' by the repair function of lung and colon injury.
