趋化因子CXCL12与其特异性受体CXCR4在胃癌组织中的表达及其与临床病理特征的关系 被引量:6

Relationship between expression of CXC chemokine receptor-4 and CXCL12 in gastric cancer and several clinicopathological factors

摘要 目的 通过检测趋化因子受体4(CXCR4)与趋化因子CXCL12在人胃癌组织中的表达,及其与胃癌临床病理特征的关系.方法 应用免疫组织化学方法检测35例正常胃组织、95例胃癌组织中CXCR4、CXCL12的表达,分析两者与临床病理特征之间的关系.结果 CXCR4蛋白在正常胃组织、胃癌组织、淋巴转移癌、肝脏转移癌、腹膜转移癌的阳性表达率分别为0(0/35)、72.6%(69/95)、89.2% (69/95)、89.4% (58/65)和100.0% (14/14);CXCL12蛋白在正常胃组织、胃癌组织、淋巴转移癌、肝脏转移癌、腹膜转移癌的阳性表达率分别为48.6%(17/35)、84.2%(72/95)、69.2%(45/65)、52.6%(10/19)、35.7% (4/14).胃癌CXCR4、CXCL12蛋白表达与肿瘤大体形态、浸润深度、淋巴管和血管的侵袭、淋巴结、肝脏和腹膜转移明显相关,而与年龄、性别、肿瘤位置和肿瘤大小无明显相关.胃癌组织中CXCR4与CXCL12蛋白表达呈正相关(r=0.509,P<0.05).结论 CXCR4和CXCL12的阳性表达与胃癌的发生、增殖、生长及转移等生物学行为密切相关. Objective To investigate the expression of chemokine CXCL12 and its receptor CXC chemokine receptor-4 (CXCR4) protein in gastric cancer and their correlation with several clinicopathological factors.Methods Tirty-five cases of normal gastric tissues and 95 cases of gastric cancer tissues were examined with immunohistochemistry to detect the expression of CXCR4 and CXCL12.The correlation between the CXCR4 and CXCL12 expression and clinicopathological factors was analyzed.Results The positive expression rate of CXCR4 protein in gastric tissue,gastric cancer tissue,lymphatic metastasis,liver metastasis and peritoneal metastasis was 0 (0/35),72.6% (69/95),89.2% (69/95),89.4%(58/65) and 100.0% (14/14),respectively.The positive expression rate of CXCL12 protein in gastric tissue,gastric cancer tissue,lymphatic metastasis,liver metastasis and peritoneal metastasis was 48.6%(17/35),84.2% (72/95),69.2% (45/65),52.6% (10/19) and 5.7% (4/14),respectively.The expression of CXCR4 and CXCL12 was correlated with the tumor general morphology,depth of invasion,tumor' s invasion on lymphatic nodes and vascular vessels and metastasis of lymph node,liver and peritoneum,but not with age,sex,location of the tumor and tumor size.The expression of CXCL12 was significantly correlated with CXCR4 (r =0.509,P 〈 0.05).Conclusion CXCR4 /CXCL12 in gastric cancer tissues presented high expression level,along with occurrence,progression and metastasis of gastric cancer.The biological axis may become an important molecular marker to cooperate with gastric cancer progression,invasion and metastasis,and indicate poor prognosis.
