摘要
运用经典试管反应法结合纸色谱法对大金发藓的化学成分进行初步测定,其中含有挥发油、油脂、糖类、皂苷、生物碱、黄酮、香豆素及其萜类化合物。采用MTT法对大金发藓醇提物进行体外细胞毒活性检测,其对多种肿瘤细胞的生长均产生一定影响,并可显著抑制L1210白血病细胞的增殖(IC5077.22μg/m L)。以三种不同类型的白血病细胞(L1210小鼠淋巴白血病细胞,K562人慢性髓系白血病细胞和U937人急性单核白血病细胞)为实验模型,比较了大金发藓孢子体和配子体细胞毒活性的差异。在相同条件下,孢子体对白血病细胞的杀伤能力远优于配子体。本文首次比较研究了大金发藓孢子体和配子体的抗癌活性,该结果不仅为大金发藓的传统用药提供了科学依据,同时也为大金发藓抗肿瘤新药研发方向提供了新思路。
Polytrichum commune L. ex Hedw is a traditional Chinese herb for treatment of fever, hemostatic, uterine pro- lapse and especially for lymphocytic leukemia. The aim of present study was to investigate the chemical components and the possible anti-prollferative activity of P. commune ethanol extract. The systematic pre-test method was used to prelimi- narily analyze the chemical components of P. commune. The obtained results showed that there were essential oils ,lipid, saccharide, glycosides, saponin, alkaloid, flavonoid, coumarin and terpene lactone in P. commune. The cytotoxlc activity was measured by MTT assay. Results showed that the cell viability of L1210 cells was reduced by P. commune ethanol extract in a concentration-dependent manner, and the ICso value was about 77.22μg/mL at 24 h post treatment. In addi- tion, the anti-tumor effects of ethanol extract of sporophyte and gametophyte from P. commune were comparatively stud- ied. M'IT assay results showed that both of them can inhibit proliferation of leukemia cells ( L1210 cells, K562 cells, U937 ceils). However,the anti-tumor activity of spomphyte was found to be higher than that of gametophyte. This was the first report on comparative research of anti-tumor effect of sporephyte and gametophyte from P. commune in cultured leukemia ceils ,which provided scientific basis for its usefulness as traditional medicine. Further studies are needed to confirm the precise mechanism not only the crude extract but as well the monomeric chemical substances of P. commune.
