华东地区部分猪场猪繁殖与呼吸综合征病毒检测及其ORF5遗传变异分析 被引量:2

摘要 为了实时掌握猪场PRRSV的感染情况,对其遗传变异趋势进行跟踪分析,本文利用RT-PCR方法对2011—2013年采自苏北地区和上海浦东地区部分猪场疑似猪繁殖与呼吸综合征(PRRS)发病猪的血清进行PRRSV检测,并对ORF5基因进行扩增测序,分析PRRSV ORF5的遗传变异情况。试验共采集样本328份,其中苏北某猪场258份,检测阳性样本为113份,浦东地区70份,阳性样本17份。每次检测选择一个样品进行ORF5测序分析。结果表明,本文所获得的15株分离株的ORF5序列与美洲型毒株氨基酸同源性为80.2%-99%,与欧洲型毒株同源性为53.4%-60.1%,均属于美洲型。进化树分析表明,有12个ORF5序列与高致病性PRRSV参考毒株同缘关系较近,其中包括苏北某猪场10份及浦东某猪场2份。结果显示,两猪场均存在PRRSV感染情况,且高致病性PRRSV与经典PRRSV并存,病毒ORF5中和表位都有变异发生,PRRSV的分布没有呈现明显的地域性差异。本试验为进一步研究华东地区猪场PRRSV感染情况及分子流行病学特征积累了有益资料。
