摘要
目的探究大鼠创伤性脑损伤(TBI)后不同时间点血管生成素(Ang)-1和Ang-2的表达情况。方法依据Marmarou方法制备脑损伤模型。在脑损伤后1 h,6 h,12 h,24 h和72 h 5个亚组中,采用改良神经功能严重程度评分系统(m NSS)进行神经功能评分,酶联免疫吸附测定(ELISA)法检测脑组织中Ang-1和Ang-2水平。结果 TBI伤后脑组织Ang-1含量1-24 h无变化,72 h显著上升;Ang-2含量1 h无变化,6 h开始显著上升,24 h达顶峰,72 h有下降但仍高于6 h组。重度脑损伤大鼠Ang-1和Ang-2水平较中度损伤大鼠升高早,幅度大。结论脑损伤后脑中Ang-1和Ang-2的表达呈现规律性变化,可能同继发性损伤和组织修复相关。
Objective To explore the expressions of angiopoietin( Ang)- 1 and- 2 in rats at different time points after traumatic brain injury( TBI). Methods The TBI model of rat was established by the method of Marmarou. In 1 h,6 h,12 h,24 h and 72 h 5 sub- groups after TBI,the modified neurologic severity score test( m NSS) was used to evaluate the neurological score and the levels of Ang- 1 and Ang- 2 in brain tissues were measured by enzyme- linked immunosorbent assay( ELISA). Results The level of Ang- 1 in brain tissue after TBI did not change from 1 h to 24 h group,but increased significantly compared with 72 h group. The level of Ang- 2 did not change in 1h and increased significantly from 6h,which reached the peak at 24 h. The level of Ang- 2 had decreased at 72 h group which was still higher than 6 h group. The level of Ang- 1 and Ang- 2 in severe brain injury group increased earlier,with large magnitude,than that of medium injury group. Conclusion The expressions of Ang- 1 and Ang- 2 after TBI showed regular changes which may be related to secondary damage and tissue repair.
