改变鼻咽癌细胞免疫原性对其迁移能力的影响

摘要 目的探讨改变鼻咽癌细胞的免疫原性对鼻咽癌迁移能力的影响。方法运用激光扫描共聚焦显微镜观察鼻咽癌细胞MHC-I、MHC-II分子的表达,划痕迁移实验检测鼻咽癌细胞的迁移能力。细胞分为四组:A组:鼻咽癌CNE-2Z细胞;B组:鼻咽癌CNE-2Z细胞+IL-2;C组:鼻咽癌CNE-2Z细胞+IFN-γ;D组:鼻咽癌CNE-2Z细胞+IL-2+IFN-γ。对各组结果进行比较分析。结果 A组鼻咽癌细胞的MHC-I、MHC-II分子几乎不表达,B组、C组和D组鼻咽癌细胞的MHC-I分子增强,MHC-II分子没有明显变化,其免疫原性增强。细胞划痕迁移实验表明,与A组比较,B组、C组和D组鼻咽癌细胞的迁移能力减弱,有统计学意义(P<0.05)。结论改变鼻咽癌细胞的免疫原性对其迁移能力有影响,使鼻咽癌细胞的免疫原性增强,其迁移的能力减弱。 Objective To investigate the changes ofnasopharyngeal carcinoma cells immunogenicity impact on the migration ability of nasopharyngeal carcinoma (NPC). Methods Laser scanning confocal microscopy was used to observe the expression of MHC-I and MHC-II molecules in nasopharyngeal carcinoma cells, scratches migration experiment was used to detect the migrational ability of nasopharyngeal carcinoma cells. Cells were divided into four groups: group A: nasopharyngeal carcinoma CNE-2Z cells; group B: nasopharyngeal carcinoma cell line CNE-2Z + IL-2; group C: nasopharyngeal carcinoma cell line CNE-2Z + IFN-γ; group D: nasopharyngeal carcinoma cell line CNE-2Z + IL-2 + IFN-γ. The results of each group were compared and analyzed. Results Group A: nasopharyngeal carcinoma cells' MHC-I, MHC-II molecules hardly expressed, group B, C and D: nasopharyngeal carcinoma cells' MHC-I molecules increased, but MHC-II molecules had no obvious changes, and its immunogenicity increased. Scratches migration experiments showed that compared with group A, nasopharyngeal carcinoma cells' migration abilities of group B, group C and group D were abate, which were statistically significant (P〈0.05). Conclusions The changes of immunogenicity of nasopharyngeal carcinoma cells has effects on the migration ability, which is the increase of nasopharyngeal carcinoma cells' immunogenicity, then the abatement of migrational ability.
