摘要
检测猪瘟疫苗免疫失败母猪血液中的CD4+、CD8+T细胞及IL-4、IFN-γ细胞因子,探讨猪瘟疫苗免疫失败的机制。随机采集规模化猪场猪瘟疫苗免疫后的母猪血液样品,用ELISA检测猪瘟抗体水平,挑选出猪瘟抗体阴性的母猪,以猪瘟抗体阳性母猪作为对照,采用ELISA、流式细胞术等方法检测分析血液中T淋巴细胞亚群及IFN-γ和IL-4。结果表明,母猪猪瘟疫苗免疫后,该场的猪瘟抗体阳性率刚能达到标准,仍有26%的猪群免疫失败。猪瘟抗体阴性组中CD4+T细胞百分数显著低于猪瘟抗体阳性组,CD3+、CD8+细胞百分数、CD4+/CD8+以及IL-4、IFN-γ浓度在阴性和阳性组中各指标差异不显著,证实母猪CD4+细胞百分数较低可能是导致猪瘟疫苗免疫失败的原因之一,为改善猪瘟疫苗免疫效果提供指导思路。
To investigate the T lymphocyte subpopulations and cytokines in sows with CSFV vaccine immunization failure. Bloodsamples were randomly collected from sows after vaccination. The CSF antibody-negative sows were screened by ELISA and CSF anti-body-positive sows as control group. ELISA and flow cytometry were used to detect T lymphocyte subpopulations and cytokines inblood. The results showed that the positive rate of vaccinated herds reached the national requirement after vaccination,but there are26% sows fail after vaciinatione. The percentage of CD4+T lymphocyte subpopulations in CSFV antibody-negative group was signifi-cantly lower than that of positive group. There was no difference between CSF antibody-negative and positive groups in the percent-age of CD3+and CD8+T lymphocytes,CD4+/CD8+ratio,IL-4 and IFN-γ levels in serum.This information should aid the improvement of CSFV vaccination.
