摘要
目的研究CD133、CD44和PSCA在肝癌组织、癌旁肝组织和肝癌细胞系中表达的特异性。方法采用免疫组化SP法检测CD133、CD44和PSCA在70例肝细胞癌(HCC)、50例肝内胆管细胞癌(ICC)组织及培养的HepG2细胞的表达。结果 CD133、CD44和PSCA在HCC组织中,阳性率分别为58.6%(41/70)、61.4%(43/70)和40%(28/70);在ICC组织中的阳性率分别为56%(28/50)、52%(26/50)和44%(22/50);统计学分析显示三者在HCC和ICC的阳性率差异不显著(χ2=0.824,P>0.05);但PSCA在ICC的阳性表达强度明显高于HCC(P<0.05)。CD133、CD44、PSCA阳性细胞在HCC、ICC和HepG2培养细胞中的分布均存在散在和成簇/片状两种形式,其中CD133以散在分布为主,而CD44和PSCA则多呈成簇/片状分布。在HCC和ICC癌的旁组织中,CD133、CD44和PSCA阳性表达率分别为51.4%(36/70)、47.1%(33/70)和32.9%(23/70)以及46%(23/50)、32%(16/50)和36%(18/50),比较3种蛋白在HCC和ICC的癌旁组织中的阳性率无明显差异(P>0.05)。CD133、CD44、PSCA在HCC、ICC及其癌旁组织中的阳性表达有明显的一致性。结论 CD133、CD44和PSCA可能均是肝癌干细胞的特异性表面标记物,其中CD133的特异性最强;而PSCA可能是ICC较为特异的癌干细胞表面标记物。
Objective To study the specificity of expression of CD133,C144 and PSCA in liver cancer,pericarcinomatous liver tissues and HepG2 cells.Methods The immunohistochemistry was used to detect the expression of CD133,CD44 and PSCA in 70 hepatocellular carcinomas (HCC),50 intrahepatic cholangiocarcinomas (ICC) and HepG2 cells.Results In the-HCCs,the positive rates of CD133,CD44,PSCA were 58.6% (41/70),61.4% (43/ 70),40% (28/70).In the ICCs,the positive rates were 56% (28/50),52% (26/50),44% (22/50),respectively.The positive expression rates of CD133,CD44 and PSCA had no significant difference in HCCs and ICCs (x2 =0.824,P > 0.05),but the intensity of positive expression of PSCA in the ICCs was significantly higher than in HCCs (P < 0.05).The distributions of CD133,CD44,PSCA-positive cells in HCC,ICC and HepG2 cells existed two forms:scattered and clusters/flaky,and the scattered was the main distribution of CD133,while the CD44 and PSCA-positive cells were mainly distributed in clusters/flaky.The positive expression rates of CD133,CD44 and PSCA were 51.4% (36/70),47.1% (33/70),32.9% (23/70) and 46% (23/50),32% (16/50),36% (18/50) in the tissues surrounding HCC and ICC respectively,and the significant difference was not found (P > 0.05) for the expression rates of three kinds of proteins in the tissues nearby HCC and ICC.They for the expression of CD133,CD44,PSCA in the HCC,ICC were obviously consistency in their pericarcinomatous liver tissues.Conclusions CD133,CD44,PSCA might be specific markers of liver carcinoma stem cells,and CD133 might be the most specific surface marker of liver carcinoma stem cells,while PSCA might be the more specific surface markers for ICC.
