法医DNA样品自动化预处理平台布局设计及应用

摘要 目的:系统介绍自行设计研发的法医DNA样品自动化预处理平台的整体布局,并检测其提取效果,为该平台的使用建立依据。方法:基于磁珠提取法结合现有提取试剂盒的基本提取步骤,对该平台的分离、吸附、洗涤、洗脱等流程进行详细介绍,并针对血斑样本进行实际检测。结果:40个血斑样本在该平台上提取的DNA,其OD260/OD280均大于1.75,扩增产物在GA118-16A遗传分析仪上进行电泳,STR分型准确,位点完整,无等位基因丢失,图谱峰形尖锐清晰,并且信号较强。结论:法医DNA样品自动化预处理平台对整体布局及运动路径进行合理设计,从而加快了DNA提取速度,提高DNA提取纯度,可满足法庭科学DNA提取的应用要求。 Objective: To design a forensic automation platform for DNA samples preprocessing and test the extraction effect. Methods: Combined with the basic extraction step of magnetic beads method, all the steps including separation, adsorption, washing and elution were carried out on the platform. Results: With our platform, the DNA samples from 40 blood samples were extracted, and the OD260/OD280 was larger than 1.75, PCR products were analyzed by electrophoresis in GA118-16 A Genetic Analyzer. The results showed that STR typing was accurate, the locus was intact, and no allelic lost. The peak was sharp and clear, and the signal was with high intensity. Conclusion: The forensic automation platform for DNA sample preprocessing could step up DNA extraction, improve the purity of extracted DNA, and was suitable for the application requirements of forensic DNA extraction.
