摘要
目的:探讨基于蛋白质组学技术的脾阴虚模型大鼠回肠组织的差异性蛋白表达。方法:SPF级SD大鼠23只,随机分为健康对照组和脾阴虚实验组。采用饮食不节加劳倦过度结合耗伤阴液的方法塑造脾阴虚证大鼠模型。经模糊数学评价方式判定后,分离出回肠组织,应用蛋白质组学技术,鉴定健康对照组与实验组回肠组织蛋白表达差异,经MASCOT搜库鉴定差异蛋白质。结果:脾阴虚实验组回肠组织发现6个差异表达蛋白点(P<0.05)。结论:脾阴虚证的发生与热休克蛋白90表达下调密切相关。
Objective:To identify ileum tissue differential proteins in spleen Yin deficiency syndrome rats. Methods:Totally 23 rats of SPF grade were randomly divided into the control group and the experimental group. Rats model of spleen Yin deficiency syndrome was established by the improper diet,overstrain and consumption of Yin fluid. The ileum tissues were separated after identified fuzzy method and the changes of expression in proteomics were identified between the control and the experimental groups by proteomics technology. The differential proteins were identified using MASCOT search library. Result:Six proteins were differentially expressed in spleen Yin deficiency experimental group( P〈0. 05). Spleen Yin deficiency syndrome was closely related with decreasing expression of heat shock protein 90.
