骨髓间充质干细胞上清液对2,5-己二酮致PC12细胞损伤的影响

摘要 目的探讨骨髓间充质干细胞上清液(BMSCs-CM)对2,5-己二酮(2,5-HD)致PC12细胞损伤的保护作用。方法体外培养、纯化大鼠骨髓间充质干细胞(BMSCs),收集第5代BMSCs的上清液。PC12细胞暴露于2,5-HD,并将大鼠分为4组,正常对照组:细胞培养体系中不加入任何药物和上清液;BMSCs-CM对照组:在细胞培养体系加入终体积分数40%的BMSCs-CM;2,5-HD损伤组:3种不同浓度(5、10、20 mmol/L)2,5-HD作用PC12细胞24 h,制备2,5-HD细胞损伤模型;BMSCs-CM保护组:20 mmol/L 2,5-HD作用细胞的同时,分别加入终体积分数10%、20%、40%的BMSCs-CM。以上各组HE染色后在光镜下观察,并利用MTT法检测细胞活性。结果 5 mmol/L 2,5-HD损伤组细胞丧失原有形态,呈圆形、椭圆形,细胞活性下降,细胞损伤程度随2,5-HD浓度的增加而下降,20 mmol/L 2,5-HD损伤组细胞损伤最严重(P<0.05)。10%BMSCs-CM保护组异常形态细胞减少,细胞活性高于20 mmol/L 2,5-HD组(P<0.05);40%BMSCs-CM保护组保护效果最好,细胞形态趋近正常,细胞活性明显优于20 mmol/L 2,5-HD组(P<0.05)。结论 2,5-HD暴露导致PC12细胞损伤,BMSCs-CM对2,5-HD致细胞损伤具有明显的保护作用。
