摘要
利用黑曲霉、米曲霉、枯草芽孢杆菌作为发酵羊胎盘菌种,筛选出黑曲霉作为发酵羊胎盘的菌株,制备羊胎盘活性肽.通过单因素试验研究了料液pH、碳源含量、发酵时间对制备羊胎盘抗氧化肽工艺的影响,在此基础上,以DPPH自由基的清除率为指标,通过响应面法优化得到了最佳发酵工艺条件为pH值4.61、葡萄糖添加量2.79%、发酵时间76.38h,此条件下的DPPH清除率为83.78%;对发酵液进行超滤处理得到6组分,其中60%的产物分子质量均<10 000u,不同组分的抗氧化活性大小为分子质量为3 000~10 000u的产物高于分子质量<3 000u的产物,并得出了发酵液中不同分子质量段的多肽含量分布曲线.
Using Aspergillus niger,Aspergillus oryzae and Bacillus subtilis as fermentation starter to ferment sheep placenta,A.niger was screened as the optimal fermentation strain to prepare sheep placenta bioactive peptide.The effect of liquid pH,carbon source content,fermentation time on the technology of sheep placenta bioactive peptide was studied.On the basis of single factor experiment,using DPPH free radical scavenging rate as index,the optimal technology was determined by response surface methodology as follows:pH 4.61,glucose addition 2.79%,fermentation time 76.38 h.Under this condition,the scavenging activity of DPPH free radicals was 83.78%.Six components were acquired by ultrafiltration of fermentation broth,among these,60% of the products were less than 10 000 u molecular weight,and the DPPH free radicals scavenging activity of components with 3 000-10 000 u molecular weight were higher than that of less than 3 000 u molecular weight.Meanwhile,the polypeptide content distribution curve of different molecular weight content in fermentation broth was acquired.
