摘要
目的研究声光可调滤光器(AOTF)近红外光谱技术,以实现淫羊藿提取过程的在线检测。方法采用AOTF近红外光谱仪,在淫羊藿提取的生产过程中实时采集近红外光谱,实时取样,采用紫外分光光度法进行离线检测。通过一阶微分PLS算法建立校正模型,利用外部验证和内部验证的方法考察模型预测的准确性。结果分别对提取过程3次煎煮提取液总黄酮含量建立模型,其相关系数分别为0.9699、0.9811、0.9059,内部验证的平均偏差为-0.28%、-0.21%、1.43%,外部验证的平均偏差分别为-0.72%、-0.26%、3.54%。结论 AOTF近红外光谱技术可有效地应用于淫羊藿提取过程的在线检测和质量控制。
Objective To study the near-infrared spectroscopy technology of acousto-optic tunable filter (AOTF) in order to achieve online detection for Epimedium brevicornum Maxim extracting process. Methods Near infrared spectrometer of AOTF was used to real-time collect near-infrared spectroscopy in production process of Epimedium brevic ornum Maxim extraction.Offline detection was conducted by real-time sampling,ultraviolet spectrophotometry.The cali-bration s was created by first-order differential PLS algorithm,the accuracy of model prediction was inspected by the method of external validation and internal validation. Results The model of total flavonoids content of 3 times decoction extract of extracting process was built respectively,and correlation coefficient was respectively 0.9699,0.9811,0.9059, the average deviations of internal validation respectively was-0.28%,-0.21%,1.43%, the average deviations of external validation respectively was -0.72%,-0.26%,3.54%. Conclusion AOTF near-infrared spectroscopy technology can be effectively applied to online detection and quality control for Epimedium brevicornum Maxim extract process.
