摘要
以不同花色、不同发育时期的鸳鸯茉莉(Brunfelsia acuminate Benth.)花瓣为材料,通过单因素试验方法,分别以不同浓度及pH值的Tris-HCl和磷酸缓冲液(PBS),获得提取花瓣中过氧化物酶(POD)的最佳浓度和pH值,并对不同花色、不同发育时期花瓣的POD活性进行测定。结果分别选出0.4 mol/L Tris-HCl,pH7.0和0.1 mol/L PBS,pH7.0 2种缓冲液组合,其中以后者的提取效果最好,并通过POD同工酶谱得以证明;不同发育时期POD活性:白花>淡紫花>紫花,花瓣颜色褪变与POD活性呈正相关,且差异均达显著水平(p<0.05);花瓣中不同形态的POD活性之间差异显著,可溶态POD活性远大于结合态的POD活性。
The petals of Brunfelsia acuminata Benth during development stages were used to determine POD activity through single-factor experiment design with different concentration and pH value of Tris-HC1 and PBS buffers in the extraction of petal POD to obtain the optimal combination. The results showed that: Among the selected buffers 0.4 mol/L Tris-HC1 (pH7.0) and 0.1 mol/L PBS (pH7.0), he latter was better, which could be confirmed by POD isoenzyme spectrum; POD activity in different development stages was as follows: white 〉 mauve 〉 purple, and POD activity was positively related with the petal color fading with an extremely significant level (p 〈0.05); The POD activity of different forms in petals varied considerably; The soluble POD activity exceeded the conjuncted one.
