11种杀虫剂对松墨天牛延伸因子2(EF-2)基因表达的影响 被引量:1

The Expression of the Elongation Factor 2 Gene from Monochamus alternatus in Response to 11 Kinds of Insecticide

摘要 [目的]研究杀虫剂对松墨天牛延伸因子2(MaEF-2)的影响,为深入研究MaEF-2的功能、利用延伸因子基因靶标有效防治松墨天牛提供参考.[方法]从松墨天牛文库中分离得到EF-2cDNA序列,利用RT-qPCR技术测定了11种杀虫剂胁迫后MaEF-2的相对表达量.[结果]得到的MaEF-2基因的部分序列,长度为1 184bp,编码287个氨基酸;与黑腹果蝇(Drosophila melanogaster)和家蚕(Bombyx mori氨基酸相似性分别为92%和93%.RT-qPCR测定结果表明,杀虫剂作用下MaEF-2均上调表达,表达量为对照的1.28 ~11.59倍.[结论]啶虫脒的相对表达量最高,毒死蜱和噻虫啉的表达量最低,这可能表明松墨天牛产生了自我保护反应. [Objective] This study provides molecular data for the influence of insecticides on M.alternatus EF-2,and the reference for the further research on the functions of M.alternatus EF-2 which can be a target against M.alternatus.[Method] A partial eDNA sequence of elongation factor 2 gene from M.alternatus library was isolated and named MaEF-2,and the relative expression of MaEF-2 in response to 11 kinds of insecticides was determined by RT-qPCR.[Result] The results showed that MaEF-2 is 1184bp in length which has 92% and 93% amino acid similarity with the EF-2 of Drosophila melanogaster and Bombyx mori,respectively.MaEF-2 were up regulated under pesticides stress,and the expression level was changed from 1.28 times to 11.59 times as that as the control.[Conclusion] The expression of acetaniprid was the highest,the expression of thiacloprid and chlorpyrifos was the lowest,all of which showed that M.alternatus produced a self-protective reaction to the insecticides.
