摘要
目的:制备紫甘薯花色苷提取物并分析其主要成分,研究其对3T3-L1前脂肪细胞生长增殖的抑制作用。方法:应用响应曲面法优化提取条件;用高效液相色谱法(HPLC)分析其主要成分;以3T3-L1前脂肪细胞为体外模型,利用MTT法得到最佳铺板密度,得到细胞生长曲线,从而在最佳密度和对数生长期检测不同浓度紫甘薯花色苷提取物对3T3-L1前脂肪细胞的增殖的影响。结果:花色苷提取物总量达到39.79 mg/g;选择细胞接板密度为5×103 cfu/mL,细胞铺板12h之后继续培养24 h,紫甘薯花色苷从最低质量浓度15.625μg/mL时即相对于空白组能显著抑制3T3-L1前脂肪细胞生长增殖的作用(P<0.05)且具有剂量效应关系,紫甘薯花色苷半数抑制浓度IC50为(413.853±2.617)μg/mL。结论:体外细胞实验显示紫甘薯花色苷具有抑制脂肪细胞生长增殖,提示紫甘薯花色苷具有预防肥胖的作用。
Objective: This study is conducted to optimizate extraction conditions of purple sweet potato anthocyanins and analysis of the content of anthocyanins in the extract, so that to study the purple sweet potato anthocyanin extract on 3T3-L1 preadipocytes inhibition of cell growth and proliferation. Methods: Using the powder of purple sweet potato as raw material, the Box-Behnken experimental design to optimizate extraction conditions such as microwave power, time, solid-liquid ratio, concentration of solvent and analysis of the content of anthocyanins in the extract by HPLC method. 3T3-L1 preadipocytes in vitro model, using MTT assay to obtain the best plated density, to obtain the cell growth curve, and thus the optimum density and the logarithmic phase of different concentrations of purple sweet potato extract anthocyanin 3T3-L1 preadipocytes proliferation. Results: The content of anthocyanins was 39.79 mg/ g, cell adapter plate density was 5×10^3 cfu/mL, cultured cells were plated 12 h after 24 h, Purple sweet potato anthocyanins from the lowest concentration 15.625 μg/mL when compared to the control group that could have a significant inhibition of 3T3-L1 preadipocytes growth and proliferation (P〈0.05), the IC50 of purple sweet potato anthocyanins was 413.853±2.617 μg/mL. Conclusion: In vitro experiments showed purple sweet potato anthocyanins inhibit fat cell growth and proliferation, suggesting purple sweet potato anthocyanins have a preventive role in obesity.
