白塞病相关新自身抗原的初步免疫学分析 被引量:3

Preliminary Immunological Analysis of Novel Autoantigen(s )in Behcet's Disease

摘要 为探讨白塞病是否存在特异性循环自身抗体,我们应用免疫印迹结合免疫沉淀技术对39例该病患者血清进行了初步筛查。结果发现,分别有8份(20.5%)、9份(23.1%)血清识别相对分子质量约为100 000、50 000的细胞内性质不明成分,其中4份标本(10.3%)与上述两种蛋白反应;在免疫沉淀分析中,15份血清(38.5%)可主要沉淀细胞中相对分子质量约为200 000、160 000、120 000、100 000的某些蛋白,部分免疫印迹阳性的血清其免疫沉淀分析也呈阳性结果。研究提示白塞病血清中可能存在作用于细胞内未知抗原的IgG型自身抗体,值得深入探索,以澄清靶抗原的性质,明确相应自身抗体对白塞病的特异性、血清学诊断价值及其可能的免疫致病机制。 Immunological abnormalities are actively involved in the disease process of Beh鏴t 's disease. in an effort to search whether there is any specific circulating autoantibody present in the disease, a preliminary study using chemilumi-nescent-immunoblot in combine with immunoprecipitation was carried out on sera derived from a cohort of 39 randomized enrolled Chinese patients with Bechet 's disease. It was found that 8 (20.5%)and 9 (23.1%)sera out of the total 39 sera recognized unknown intracellular components with molecular mass around 100 kD and 50 kD respectively, and 4 (10.3%)of them showed reactivity to both proteins as determined by immunoblot. Furthermore, 15 sera (38.5%)could mainly immunoprecipitate certain cellular proteins at 200, 160, 120, 100 kD. In comparison, part of the immunoreac-tive sera in immunoblot demonstrated positive immunoprecipitation results. This study suggested that some autoantibody (ies )(IgG class )targeting at yet uncharacterized cellular antigen (s ) may exist in Behcet 's disease sera, which de-serves further study to decipher the identity of the target antigen (s ) , clarify the specificity of the corresponding autoantibody (ies ) for Behcet 's disease and potential serodiagnostic usefulness, possible immunopathogenetic significance thereof.
