抗禽流感病毒H_5亚型血凝素特异性单克隆抗体的研制 被引量:21

摘要 以禽流感H5 亚型病毒免疫Balb/c小鼠 ,取其脾细胞与SP2/0骨髓瘤细胞融合 ,用血凝抑制试验检测细胞培养上清 ,结果获得3株阳性细胞株 ,分别命名为4B6、4A3、3H1。经2次亚克隆后 ,100 %杂交瘤细胞保持了分泌抗禽流感病毒H5 亚型抗体的能力。实验证明 ,所有这3株单克隆抗体仅与试验的H5 病毒株反应 ,而不能与禽流感H9 亚型病毒、鸡新城疫病毒、鹅新城疫病毒和鹅腺病毒等反应。间接免疫荧光试验检测结果表明这3株单克隆细胞的培养上清均能与感染H5 亚型病毒的成纤维细胞呈现亮绿色荧光。 Three monoclonal antibodies to subtype H5 avian influenza virus(AIV-H5) were developed by fusing SP2/0 myeloma cells with spleen cells of Balb/c mice immunized with AIV-H5,which were designated as 4B6,4A3,and 3H1 respectively.By hemagglutination inhibition test (HI),these three monoclonal antibodies reacted specifically with AIV-H5 but not with AIV-H9,Newcasttle disease virus in chickens (NDV),NDV in geese,advenovirus in geese.Chicken embryo fibroblast (CEF) cells infected with AIV-H5 could be detected by these monoclonal antibodies in indirect immunofluorescent assay (IFA).The results suggested that these monoclonal antibodies will be very useful in diagnosing avian influenza.
