摘要
目的 :观察中药调肝导浊汤对家兔实验性AS模型肝脏脂代谢及体外培养肝细胞膜脂蛋白受体的影响 ,探讨中药防治AS的机制。方法 :高胆固醇饮食饲喂家兔 10W造成AS模型 ,给药 6W结束实验 ,取肝脏匀浆测定TC、TG变化。此外 ,体外培养大鼠肝细胞 ,原位杂交法计数膜LDL受体阳性表达。结果 :调肝导浊汤可有效降低肝组织脂质含量 ,中药可以显著增强肝细胞膜LDL受体mRNA表达。结论 :调肝导浊汤具有良好的保护肝脏、调整脂代谢的作用 ,从而防治AS。
Objective:To understand the important effect of the liver on the metabolism of lipid from the hyperlipemia model of rabbit and the influence of lipoprotein receptor in the cultured hepatocyte membrane of the tiaogandaozhou decoction,observe the action of preventing the formation and progress of atherosclerosis of it.Method:The rabbits were fed with heavy dosage of cholestorl for 10 weeks and formalized the model of atherosclerosis.The drug were given for 6 weeks.After the experiment ended,put the object to death and sample the liver,then assay the changes of the cholestorl and triacylglycerol.Besides,the hepatocyte of rat was cultured and analysis was made on the mRNA expression of low density lipoprotein receptor with hybridization in vitro. Result:The contents of the lipid in the liver were lowered obviously in the Chinese medicine group.;The mRNA expression of low density lipoprotein receptor in the hepatocyte membrane was strengthened obviously in the Chinese medicine group.Conclusion:The Chinese drugs has a good effect on protecting the liver and regulating the metabolism of lipid.which can pevent the formation and progress of atherosclerosis.
