摘要
目的 :研究细胞内Ca2 + 信号在肾阳虚发病中的作用及补肾中药的调整作用。方法 :用流式细胞仪及钙荧光探针Flou 3 AM对肾阳虚大鼠下丘脑 垂体 肾上腺轴、血淋巴细胞胞内Ca2 +([Ca2 + ]i)和血清钙进行测定。结果 :肾阳虚大鼠下丘脑和血淋巴细胞 [Ca2 + ]i及血清钙显著升高 ,补肾中药能够显著抑制下丘脑、血淋巴细胞内 [Ca2 + ]i和血清钙的升高。结论 :肾阳虚时下丘脑、血淋巴细胞 [Ca2 + ]i和血清钙的显著升高 ,导致Ca2 + 信号的异常 ,使下丘脑 垂体 肾上腺轴功能紊乱 ,免疫功能低下 ,出现肾阳虚的症状 ,补肾中药可能通过调整下丘脑、血淋巴细胞 [Ca2 +
Objective:Study the effects of cytoplasmic Ca 2+ signal in kidney yang deficiency rat and regulation of tonify kidney herb.Methods:[Ca 2+ ]i of hypothalamus pituitary adrenal axis and lymphocyte of blood in rats were tested with flow cytometry by using [Ca 2+ ]i fluorescent probe Flou 3/AM,as well as serum calcium.Results:[Ca 2+ ]i of hypothalamus and lymphocyte of blood and serum calcium increased significantly in kidney yang deficiency rats.Tonifying kidney herb decreased [Ca 2+ ]i of hypothalamus and lymphocyte of blood and serum calcium.Conclusion:[Ca 2+ ]i of hypothalamus and lymphocyte of blood and serum calcium increased significantly during kidney yang deficiency,which induced abnormality of Ca 2+ signal.The change of Ca 2+ signal resulted the distrubance of function in hypothalamus pituitary adrenal axis and declination of immunity,and then the occurrence of symptom in kidney yang deficiency.Tonifying kidney herb probably exerted effect by regulating Ca 2+ of hypothalamus and lymphocyte of blood and serum calcium.
