摘要
已知乙型肝炎患者外周血单个核细胞(PBMC)中可有HBVDNA存在,但对HBV基因在PBMC内是否表达及如何表达,至今所知尚少.本文用完整PBMC涂片,PAP染色等方法研究了血清中HBV标志阳性者PRMC内HBsAg-HBV基因表达产物.实验结果表明,27例受试者中13例PBMC内HBsAg阳性.对阳性者中的4例,进一步分析HBsAg在单核细胞、T细胞及非T细胞内的分布,结果有2例HBsAg同时存在于T细胞和非T细胞内,另2例分别仅在非T细胞或单核细胞内检出.本文发现HBsAg仅存在于PBMC的胞浆中,并可分为两种染色类型:弥漫型及局灶型.提示PBMC可受HBV感染,HBV基因也可在PBMC内表达.
The presence of HBV DNA in peripheral blood mononuclear cells(PBMC) has been reported, but few detailed information dealt with the expression of HBV genome in PBMC. This paper, reports the use of unlabeled PAP method with smears of intact PBMC in research of the expression of HBV surface antigen gene in PBMC of patients with positive HBV markers in serum. In 13 out of 27 patients HBsAg detection in PBMC gave positive results. And 4 out of these 13 cases were further identified for the presence of HBsAg in monocyte, T cell, and non-T cells respectively. Two of them were HBsAg positive both in T cell and non-T cells; another one only in non-T cells, and the remaining one only in monocytes. HBsAg was present only in the cytoplasm of PBMC and appeared in two staining models, i. e. diffused and localized. The results suggest that HBsAg could be detected in T cell and non-T cells, PBMC could be infected by HBV virus and HBV surface antigen gene might be able to find its expression in these cells.
