摘要
目的 :探讨MTA1基因与胃癌浸润和转移关系。方法 :建立测定MTA1基因mRNA表达的逆转录聚合酶链反应 (RT PCR)最适条件 ,在半定量水平测定 37例胃癌标本癌组织和癌旁正常黏膜组织MTA1基因mRNA表达的相对水平 ,分析与临床和组织病理学特征的关系。结果 :37例胃癌中有 14例MTA1基因mRNA过度表达 (癌组织与癌旁正常黏膜组织MTA1基因mRNA表达半定量之比≥ 2 ) ,过度表达MTA1基因mRNA的胃癌浸润层次深、淋巴结转移率高 ,与对照组比较差异均有显著性 (P =0 .0 0 16和P =0 .0 15 2 ) ,过度表达组脉管累及、TNM分期、分化程度与剩余病例组比较差异均无显著性 (P >0 .0 5 )。结论
Objective:To study the relation between metastasis associated MTA1 gene and gastric carcinoma progression.Methods:Optimal conditions of reverse transcription polymerase chain reaction(RT PCR) were found out;then the expression of MTA1 gene mRNA in 37 gastric carcinoma samples was compared with that in corresponding normal mucosa tissue at semi quantitative level and the results were compared with clinical pathologic data.Results:A relative overexpression of MTA1 gene mRNA(tumor/normal ratio≥2) was observed in 14 of the 37 gastric carcinoma samples.Clinico pathologic correlations demonstrated that overexpression of MTA1 gene mRNA exhibited a significantly deeper wall invasion( P =0.001 6) and a higher rate of metastasis to lymph nodes( P =0.015 2) than the remaining samples;while the difference in vascular involvement,TNM staging and differentiated grading was not significant( P >0.05).Conclusions:Our data suggest that overexpression of the MTA1 gene correlates with invasion and metastasis of gastric carcinomas.
