摘要
目的 对恶性疟原虫海南株的 MSP2基因进行分型和多态性分析。 方法 采用套式 PCR法特异扩增 MSP2基因的中间可变区 (第 3区 ) ,对 4 1株恶性疟原虫分离株进行基因分型 ,并对部分目的基因片段进行直接测序。 结果 4 1份血样中有 39份共扩增得到 5 5个目的基因片段 ,以 3D7型为主 (2 8份血样 ,36个基因片段 ) ,两种等位基因家族混合感染的情况极少见。序列分析结果表明 ,海南省恶性疟原虫虫株的 MSP2中间重复序列区变异程度大。 结论 海南省恶性疟原虫的 MSP2以 3D7等位基因家族为主 。
Objective To determine the genotype distribution and to analyze the allele polymorphism of merozoite surface protein 2 (MSP2) of Plasmodium falciparum isolates in Hainan Province, China. Methods Nested PCR technique was applied to amplify the central polymorphic region of MSP2 allele for genotyping of P. falciparum . Some gene fragments were sequenced for the analysis of the allele polymorphism. Results 55 gene fragments were obtained from 39 positive blood samples, with 3D7 type alleles being predominated (36 gene fragments from 28 blood samples). The mixed infection of different allele families was rare. Sequence analysis showed that variation in the central repeat regions was remarkable. Conclusion 3D7 type allele was the predominant MSP2 allele and extensive polymorphism of the MSP2 was found in P. falciparum isolates from Hainan Province, China.
