摘要
将香菇 (子实体 )经水浸提液浸提 ,再经减压浓缩、Sevag法去除游离蛋白、乙醇沉淀得到香菇多糖蛋白粗品。通过正交实验分别得到制备浸提液的最优工艺条件 :温度85℃ ,时间 5h,加水量 1 50 ml;除蛋白的最优工艺条件 :样液∶氯仿 -正丁醇 (体积比 ) =3 ,氯仿∶正丁醇 (体积比 ) =5;而乙醇体积分数为 70 %时可沉淀出较多的糖蛋白。经过纯度鉴定 ,可得到纯度为 86.3
The extracted polysaccharide is a glycoprotein. Glycoprotein is gained by lixiviation, concentrating, sevag reagent's getting rid of free protein and precipitating through ethanol. The orthogonal experiment shows that the best lixiviating process is temperature 85℃, time 5h, water capacity 150ml, the volume ratio of glycoprotein solution to sevag reagent is 3 and sevag reagent is 5 between chloroform and butanol, this is the best sevag reagent's getting rid of free proteins process resulting from orthogonal expriment, when the alcohol consistency comes up to 70% in the gylcoprotein solution.The glycoprotein is precipitated out more than under other alcohol consistency. The semifinised product's purity is 86.3%.
