碘对Graves病患者甲状腺上皮细胞HLA-DR表达的影响 被引量:1

摘要 目的 探讨碘在Graves病 (GD)发病中的作用。方法 取手术切除的GD患者甲状腺组织及甲状腺腺瘤患者腺瘤旁正常甲状腺组织 ,进行细胞培养 ,培养液中含不同浓度的NaⅠ (0~ 1mg/L)。采用免疫印迹技术 ,观察NaⅠ对甲状腺上皮细胞 (TEC)HLA DR表达的影响 ,并以扫描电镜观察NaⅠ处理后TEC的形态学改变。结果  (1)GDTEC :培养液中含NaⅠ浓度超过 10 0 μg/LHLA DR表达增加 (P <0 .0 1) ,其增加量与NaⅠ浓度无剂量依赖关系。扫描电镜显示培养液中NaⅠ浓度超过 10 0 μg/L于加碘 12h出现甲状腺细胞膜损伤。 (2 )腺瘤旁正常TEC :NaⅠ不同浓度组均无HLA DR表达。NaⅠ浓度超过 10 0 μg/L时 ,扫描电镜未显示TEC有细胞膜的损伤。 结论 过多的碘不仅造成GD甲状腺细胞膜的直接损伤 ,且可以上调TECHLA Objective To investigate the effects of iodine on the pathogenesis of Graves′ disease (GD). Methods The thyroid epithelium cells (TEC) from GD patients and para adenoma normal tissue from thyroid adenoma patients wereculturedrespectivelywithdifferentNaⅠconcentrations from 0 to 1 mg/L. The HLA DR expression of TEC was detected with Western blot. Meanwhile, the morphological change of TEC was observed after treatment of NaⅠ by scanning electronmicroscope (SEM). Results (1) When the concentration of NaⅠ in culture medium was over 100 μg/L, the amount of HLA DR expression of TEC increased in GD group (P<0.01), but there was not related to the concentration of NaⅠ. SEM showed that the membrane of TEC was injured when TEC were cultured with over 100 μg/L NaⅠ for 12 h; (2) In para adenoma normal tissue, HLA DR was not expressed in different NaⅠ concentrations. SEM showed that TEC did not have membrane damage. Conclusion The excessive iodine not only results in the direct damage of TEC membrane, but also up regulates HLA DR expression of TEC from GD.
