谷氨酸单钠对新生小鼠视网膜神经细胞c-fos表达的影响 被引量:1

摘要 目的 观察谷氨酸对小鼠视网膜神经细胞 c- fos表达的变化 ,探讨谷氨酸的神经毒性作用 .方法 昆明系小鼠妊娠晚期经口一次性大剂量谷氨酸钠 ig(4.0 g·kg- 1 ) ,仔鼠于出生后 10 ,2 0和 30 d取眼球 ,多聚甲醛固定 ,恒冷切片 ,应用免疫组织化学方法对视网膜神经细胞 c- fos的表达进行了观测 .结果  10 ,2 0和 30日龄谷氨酸组小鼠视网膜神经细胞 c- fos表达阳性神经元分别为 14 74 ,12 5 0及 85 9cells·mm- 2 ,比正常小鼠 c- fos的表达明显增高 (14 74 vs74 4 cells·mm- 2 ,P<0 .0 5 ;12 5 0 vs5 38cells·mm- 2 ,P<0 .0 5 ;85 9vs4 0 4 cells· mm- 2 ,P<0 .0 5 ) .结论 外源性谷氨酸作用于成长发育中仔鼠的视网膜神经细胞 ,并引起细胞内 c- fos过度表达 。 AIM To observe the toxic effect of monosodium glutamate on the expression of c fos in the retinal neurons inmice. METHODS Kunming mice were given 4.0 g·kg -1 monosodium glutamate via oral administration at the late stage of pregnancy. The eyeballs of filial mice were removed on the postnatal days of 10, 20 and 30, the specimens then fixed with paraformaldehyde and sectioned into pathological slices by the Cryocut. Immunohistochemistry was used to detect the expression of c fos in retinal neurons. RESULTS In the monosodium glutamate treated mice, there were 1474 cells·mm -2 positive immunoreactive neurons of c fos on postnatal day 10, 1250 cells·mm -2 on postnatal day 20, 859 cells·mm -2 on postnatal day 30 while there were respectively 744, 538, 404 cells·mm -2 in the normal mice retinas (1474 vs 744 cells·mm -2 , P <0.05; 1250 vs 538 cells·mm -2 , P <0.05; 859 vs 404 cells·mm -2 , P <0.05). CONCLUSION Exogenous monosodium glutamate has the effects on the developing retinal neurons that it can produce an overexprssion of c fos in the retinal cells, corresponding with induction of neuronal apoptosis.
