摘要
目的 研究配制的质控血清的稳定性和影响因素。方法 将配制的HIV抗体质控血清进行不同的处理,然后统一用 3种试剂进行检测:(1)在 37℃,室温和 4℃保存不同时间;(2)加入 0.1%的叠氮钠和 0.01%的硫柳汞保存不同时间;(3)室温和-20℃之间反复冻融。结果 人工配制的质控血清在4℃条件下6个月内可以保持稳定,在室温条件下1个月内保持稳定。反复冻融及加入0.1%的叠氮钠或0.01%的硫柳汞对检测结果没有影响。双抗原夹心法试剂检测的S/CO值显著高于间接法试剂。结论 在常规条件下血清中的HIV抗体能够在较长时间内保持稳定。
Objective Study the stability of artificially prepared HIV antibody quality control serum. Methods The artificially prepared quality control serum was handled with different methods, and tested with 3 kinds of HIV antibody kits: 1)Keep in 37℃ , 4X3 or room temperature.2) add the 0.1% Sodium azide or 0.01 % Merthiolate.3)freeze and thaw 10 or 20 times. Results The quality control serum was stable at 4X3 in 6 months and at room temperature in 1 month. The test results were unchanged after the repeated freeze and thaw or the addition of preservatives.The value of S/CO were significantly higher by the sandwich reagent than by indirect reagent.Conclusion The HIV antibody was stable for a long time under the routine conditions.
