钠摄入量对心力衰竭大鼠心脏肾素-血管紧张素系统与心钠素的影响 被引量:11

摘要 目的 观察钠摄入量对心力衰竭大鼠心脏局部肾素-血管紧张素系统与心钠素的影响 .方法 经动 -静脉分流术造成大鼠充血性心衰模型 ,分为心衰组、心衰限钠组、心衰补钠组 ,假手术大鼠为对照组 ,用放射免疫分析法和原位杂交技术分别测定各组血浆和心肌血管紧张素 、心钠素含量及心肌血管紧张素原 m RNA表达水平 (吸光度 A值 ) ,同时检测心功能 .结果 心衰限钠组心房和心室血管紧张素 含量(2 0 .1± 4 .5 )和 (2 7.3± 5 .9) ng· g- 1、血管紧张素原 m RNA表达 (6 .4± 1.2 )和 (12 .6± 2 .3)显著高于心衰组 (17.5± 3.6 )和 (2 0 .1± 3.7) ng· g- 1 ,(6 .2± 1.9)和 (8.6± 1.7) (P<0 .0 5 ,P<0 .0 1) ,心室心钠素 (42 3± 6 8) ng· g- 1 也显著高于心衰组 (337± 86 ) ng· g- 1 (P<0 .0 5 ) ,心房心钠素 (6 7± 19)μg· g- 1显著低于心衰组 (85± 15 ) μg· g- 1 (P<0 .0 5 ) ;心衰补钠组心房和心室血管紧张素原 m RNA表达水平、心室血管紧张素 和心钠素与心衰组无显著差别 ,心房血管紧张素 与对照组无显著差别 ,心房心钠素 (10 1± 17) μg· g- 1 显著高于心衰组 (P<0 .0 1) .结论 心衰时不同钠摄入量可通过改变心脏局部肾素 AIM To observe effects of dietary sodium intake on cardiac renin angiotensin system (RAS) and atrial natriuretic peptide (ANP) in rats with congestive heart failure (CHF). METHODS CHF rats induced by aortocaval shunt were divided into CHF group, sodium restricted group, and sodium supplemented group with sham operation rats as control group. Radioimmunassay and in situ hybrdization techniques were used to determine the plasma and myocardium angiotensin Ⅱ (AngⅡ) and ANP contents and the expression of angiotensinogen mRNA, respectively, meanwhile cardiac function was measured. RESULTS In sodium restricted group, the atrial and ventricular AngⅡ contents [(20.1± 4.5) ng·g -1 , (27.3±5.9) ng·g -1 ], angiotensinogen mRNA expression absorbance [(6.4±1.2) , (12.6± 2.3)] were significantly higher than those in CHF group [(17.5±3.6) ng·g -1 ,(20.1±3.7) ng·g -1 and (6.2± 1.9), (8.6± 1.7)] ( P<0.05, P <0.01), the ventricular ANP contents [(423±68) ng·g -1 ] were also obviously higher than those in CHF group [(337±86) ng·g -1 ] ( P <0.05), while the atrial ANP [(67±19) μg·g -1 ] were obviously lower than that in CHF group [(85±15) μg·g -1 ] ( P < 0.05); In sodium supplemented group, the atrial and ventricular angiotensinogen mRNA expression, ventricular AngⅡ and ANP contents had no obvious change compared with the CHF group, the atrial AngⅡ had no difference as compared with the control,while the atrial ANP contents [(101±17) μg·g -1 ] were significantly higher than those in CHF group ( P <0.01). CONCLUSION Dietary sodium intake can play an important role in the development of CHF by influencing the balance of local RAS and ANP,and an appropriate supplement of sodium intake has a beneficial effect on CHF.
