白细胞介素—12和白细胞介素—18质粒对HBcAg DNA疫苗诱导小鼠(H—2^d)体液免疫应答的影响 被引量:8

Effects of the Plasmids Expressing Interleukin 12 and 18 on Humoral Immune Responses in H- 2d Mice Induced by DNA Immunization of HBV Core Gene

摘要 目的:观察编码鼠白细胞介素(IL)-12和IL-18的真核表达载体犤pcDNA3.1/IL-12(以下简称IL-12质粒)和pcDNA3.1/IL-18(以下简称IL-18质粒)犦对HBcAgDNA疫苗(pJW4303/HBc)诱导Balb/c(H-2d)小鼠免疫应答的影响。方法:肌肉注射接种IL-12质粒、IL-18质粒和HBcAgDNA疫苗,ELISA法检测小鼠血清抗HBcIgG亚类(IgG1、IgG2a)水平。结果:免疫6周后,联合注射IL-12、IL-18和IL-12+IL-18组小鼠血清的抗HBc终点滴度均明显高于单纯注射HBcAgDNA疫苗组小鼠(P<0.05),抗HBcIgG亚类以IgG2a占优。结论:IL-12、IL-18以及IL-12+IL-18联合HBcAgDNA疫苗注射能够提高小鼠血清中抗HBc水平。 Objective:To observe effects of the plasmids expressing interleukin 12 and interleukin 18 on the humoral immune responses in Balb/c(H 2d)mice induced by DNA immunization of HBV core gene. Methods:The DNA vaccine of HBV core gene,the plasmids expressing interleukin 12 and interleukin 18 were simultaneously immunized by intramuscular injection in Balb/c mice.The levels of anti HBc antibody and its isotypes (IgG1,IgG2a)were detected by ELISA methods.Results: Six weeks after the 1st immunization, the levels of anti HBc antibody in groups of mice immunized with HBV core DNA vaccine plus the plasmids expressing interleukin 12 or interleukin 18 or both interleukin 12 and interleukin 18 were all much hugher than those in groups of mice immunized with HBV core DNA vaccine alone(P< 0.05).In analysis of IgG subgroups the IgG2a was generally higher than IgG1 in all groups of mice immunized with DNA vaccines mentioned aboue. Conclusion:The plasmids expressing interleukin 12 and interleukin 18 may enhance the specific humoral immune responses induced by immunization with HBV core DNA vaccine.
