P-170糖蛋白在多药耐药(MDR)K562/ADM细胞中的定位研究 被引量:1

摘要 目的 :确定P - 170糖蛋白在多药耐药 (MDR)K5 6 2 /ADM细胞中表达的位置。方法 :通过免疫反应并利用流式细胞仪 (FCM )检测表达P - 170糖蛋白细胞的百分含量 ;用免疫电镜技术检测P- 170糖蛋白在细胞中的位置及其表达水平。结果 :敏感株K5 6 2细胞与耐药株K5 6 2 /ADM细胞对P - 170糖蛋白的表达有非常明显的差异 ;不同的K5 6 2 /ADM细胞表达P - 170糖蛋白的量差别较大 ;少量的K5 6 2细胞也有极少的P - 170糖蛋白的表达 ;细胞表达的P - 170糖蛋白位于细胞膜上。结论 :多药耐药K5 6 2 /ADM细胞表达的P - 170糖蛋白最终被镶嵌在细胞膜上 ;在该蛋白质从被表达、修饰和转运到细胞膜上的过程中 ,该蛋白质不具有成熟P - Objective: To confirm the position of P-glycoprotein in K562/ADM cells with immune electron microscope(IEM) technology. Method: The percent of K562/ADM cells of expressing P-glycoprotein was mensurated by Flow cytometer (FCM); The position and expressing level of P-glycoprotein in K562/ADM cells were confirmed with IEM. Results: There are obvious differentiation of expressing level of P-glycoprotein in K562 and K562/ADM cells, K562/ADM cell line is higher than K562. Every cell of K562/ADM expresses P-glycoprotein with different quantity, but a few of K562 cells expressed a little P-glycoprotein. The P-glycoprotein is incrusted in cell membrane finally. Conclusions: P-glycoprotein is incrusted in cell membrane of K562 and K562/ADM cells.
