摘要
对侧耳属 18个种 5 2个菌株及 3个其它属的菌株的 2 8SrDNA 5’端进行PCR扩增 ,得到长度约为 1.4 6kb的片段。对该片段分别用 7种限制性内切酶AluⅠ、BamHⅠ、HaeⅢ、HhaⅠ、HinfⅠ、MspⅠ、TaqⅠ酶切 ,结果表明 ,MspⅠ酶切片段多态性最高 ,AluⅠ对侧耳属无酶切多态性。聚类分析结果表明 ,菌株间的相似系数为 0 .5 6 9~ 1.0 0 0 ,在 92 %相似水平可将侧耳分为 5大类 :Ⅰ .红平菇和桃红侧耳 ;Ⅱ .鲍鱼菇和囊盖侧耳 ;Ⅲ .具核侧耳 ;Ⅳ .金顶侧耳 ;Ⅴ .
S rDNA 5'half of eighteen Pleurotus taxa (52 isolates) and 3 other genera as outgroups was amplified using PCR and then digested with seven restriction endonucleases. A single product 1.46 kb in size resulted from PCR. RFLP results showed that high polymorphisms were observed for MspⅠand no polymorphisms for AluⅠin Pleurotus . Dendrogram showed that five groups were distinguishable for all tested Pleurotus isolates, i.e. P. djamor & P. salmoneostramineus;P. tuber regium;P. citrinopileatus;P. abalonus & P.cystidiosus and other isolates ( P. ostreatus,P. florida,P. sapidus,P. corticatus,P. cornucopiae,P. columbinus,P. spodoleucus,P. nebrodensis,P. ferulae, P. pulmonarius & P.sajor caju,P. erygii and Pleurotus sp.).
