摘要
目的 :观察大鼠肝胆代谢功能及雌激素对胆汁分泌的反应。方法 :将 2 0只近交系Wistar大鼠随机分组 ,试验组肌肉注射苯甲酸雌二醇 ,由丙三醇稀释 ,5mg/kg体重 ,连续注射 5d。对照组肌肉注射丙三醇。所有动物于第 6天手术装置急性胆总管瘘管 ,连续收集 4次胆汁 ,前 3次每隔 2 0min收集 1次 ,第 4次收集 1h。于胆汁收集前后采血 ,测定血浆与胆汁中总胆酸 (TBA)、甘氨胆酸 (GC)、总胆红素 (TBIL)、丙氨酸转氨酶 (ALT)、天冬酸转氨酶 (AST)、碱性磷酸酶 (ALKP)、γ 谷氨酰转肽酶 (GGT)、总胆固醇(CHO)等肝胆功能指标及激素水平 ,作统计分析。结果 :大鼠胆汁中TBA、GC、TBIL、GGT显著高于血浆中浓度 ,而血浆中ALT、AST、ALKPCHOL显著高于胆汁中浓度。胆汁中TBA、GC极高 ,而CHOL、ALT、AST、ALKP、雌二醇 (E2 )甚微。雌激素注射可使胆汁分泌量显著减少 ,胆流速度显著减慢。雌激素注射后 ,血浆中CHOL显著下降 ,ALKP、GGT、E2 显著升高 ,胆汁中TBA显著下降 ,TBIL、ALKP、GGT、CHO显著升高 ,其它指标变化不大。结论 :雌激素可降低胆汁分泌功能 ,引起肝内淤胆 ,致血浆和胆汁中GGT、ALKP、E2 的升高 ,但不直接损伤肝细胞。雌激素可导致CHOL肝内合成减少而胆内排泄加快 ,但不能引起血液和胆汁中胆酸浓度发生变化。
Objective:In order to observe liver and gallbladder function in rats and how estrogen affects bile secretion. Methods: Twenty inbred Wistar rats,were divided randomly into two groups. Treat groups were injected i.m benzogynestryl dissolved in propyl alcohol, 5 mg/kg body weight,for five days. Control groups were injected i.m propyl alcohol. All animals were each planted a catheter of general bile duct in the sixth day after infusion. Bile was continuously collected four times, with an interval of 20 minutes for the first three times and one hour for the fourth time. Blood was collected before bile was gathered, and plasma was separated. TBA, GC, TBIL, ALT, AST, ALKP, GGT, CHOL and E 2 were determined in bile and plasma. The data were analyzed statistically. Results: TBA, GC, TBIL, GGT in the bile are higher than those in the plasma and ALT?AST?ALKP CHOL lower significantly in the rats. Biliary TBA,GC were very high and CHOL?ALT?AST?ALKP?E 2 were very low. After the rats were injected benzogynestryl, bile secretion values and speed decreased significantly. Compared with those of control groups, plasma ALKP?GGT?E 2 of treat groups increased and CHOL decreased significantly, biliary TBIL?ALKP?GGT?CHOL increased and TBA decreased significantly. Conclusions: Estrogen can decrease bile secretion, raise intrahepatic cholestasis, and increase GGT?ALKP?E 2 in plasma and bile, but do not damage liver cells directly. Estrogen can cause less CHOL synthesis, but does not change the concentration of plasma cholic acid.
