摘要
目的 :通过对兔骨骺软骨组织的冷冻保存和进行同种异体移植模拟研究 ,为临床骨骺软骨的保存和应用提供理论依据。 方法 :选用 1个月龄的新西兰大白兔的髂骨骨骺 2 4份。用 10 %二甲亚砜 (DMSO)冷冻保护液保护后 ,用温度速降仪使骨骺软骨的环境温度逐渐降至 - 80℃ ,然后迅速放入液氮中保存 1、3、6周。取出后在室温下置入等渗盐水中复温。其中 12份进行组织学和组织化学检查 ;另 12份作为供体 ,植入同种成年兔股骨头部分缺损中 ,分别观察 4、12和 2 4周。然后取出股骨头进行大体、组织学和组织化学检查。 结果 :实验显示 ,在上述冷冻条件下 ,大部分骨骺可存活并保存骨骺组织生长塑形 ,逐渐骨化和替代关节软骨的功能。 结论 :只要冷冻条件合适 ,骨骺软骨组织保存一定时间后仍具有生理功能 ,这为临床上修复骨骺损伤和关节软骨缺损提供了一条新的途径。
Objectives: Cryopreservation of epiphyseal cartilage is investigated in conjunction with freeze preservation and transplantation of epiphyseal allograft. Methods:Twenty four rabbits epiphyseal cartilage(0.5 cm×0.3 cm×0.15 cm) of iliac crest were exposed in 10% dimethyl sulfoxide, and frozen at 2℃/min in a control rate freezing apparatus, and stored in liquid nitrogen for 1,3 and 6 weeks respectively. They were then quick thawed in normal saline. Twelve of 24 epiphyseal cartilage were implanted into the fresh defect in the femoral head of randomly selected recipients for 1 to 6 months. Twenty four cryopreservative epiphysis were studied macroscopically and histologically. Results:Cryopreservative epiphysis showed normal structure in gross and microscopic section, and eight of twelve epiphyseal allografts revealed good remodeling in the joint with defect, the ossification process of the epiphyseal allografts seemed to be normal, the most superficial part of the graft was similar to the articular cartilage of the femoral head. Conclusions:Epiphyseal cartilage is functional physiologically after exposure to cryopreservation.
