用免疫组化及原位杂交方法探测人胚胎卵巢褪黑素受体 被引量:1

摘要 目的 :在前期工作基础上进一步观察人胚胎卵巢组织中褪黑素 (Mel)受体的存在及分布。方法 :取人胚胎卵巢 ,石蜡包埋、切片 ,用原位杂交及免疫组化方法进行检测。结果 :免疫组化方法显示卵巢细胞的细胞膜、胞浆、细胞核中Mel1a、Mel1b均呈棕褐色 ;原位杂交方法显示褪黑素受体Mel1a、Mel1b位于卵巢细胞的细胞浆及细胞核内 ,呈蓝紫色颗粒。结论 :人胚胎卵巢组织中存在有Mel1a及Mel1b两种褪黑素受体 ,位于卵巢细胞的细胞膜、胞浆、细胞核中 ,证明卵巢是褪黑素作用的靶器官 ,褪黑素可能直接作用于卵巢。 Objective: To study the distribution of melatonin receptors in embryos ovary of human.Methods:The ovaries were collected and sliced up,and stained with immunohistochemistry and in situ hybridization.Results:The membrane, plasma,and melatonin receptors Mel 1a and Mel 1b of nuclear in ovary tissue were positively dyed as brown granules with Immunohistochemistry,while all the above tissues were positively dyed as blue granules with in situ Hybridization.Conclusion:There exists melatonin receptor in ovary of human embryo,which is located in the membrane,plasma,and nuclear.It is proved that melatonin can act on the ovary and regulate the development or function of embryo ovary.
