摘要
本文报道低速离心、地塞米松和二甲亚砜对人类巨细胞病毒AD_(169)株在人胚肺细胞上增殖的影响。结果表明本法能使人类巨细胞病毒的细胞病变分别提前2~3天。地塞米松能增加人类巨细胞病毒AD_(169)株在人胚肺细胞上空斑形成能力约3倍。
The effect of low-speed centrifugation, the drugs dexamethasone and dimethyl sulfoxide on the duplication of human cytomegalovirus (HCMV) AD169 strain in the human embryonic lung (HEL) cells was studied.The results showed that cytopathic effects (CPE) of HCMV in cell cultures treated with above-mentioned factors took place 2-3 days earlier than that of the control.Preliminary application of these modified culture methods for HCMV isolation from clinical specimen revealed that among 7 cases of positive HCMV isolation obtained by dexamethasone treatment on cell culture typical CPE of HCMV arose earlier in 6 cases than that in corresponding controls.The influence of dexamethasone and dimethyl sulfoxide on the HCMV-host cell interaction was detected with HCMV plaque assay.The numbers of plaque in the treated HEL cell cultures increased by 2 to 3-fold than that in controls.The sensitivity of HEL cell cultures to HCMV infection was also enhanced by treatment with dexamethasone or dimethyl sulfoxide.In addition, strain 401 was identified to be cytomegalovirus by the plaque reduction neutralization assay.
