摘要
目的 :对实验性高脂血症大鼠血液流变学影响及其对大鼠的胚胎毒性和致畸作用。方法 :①用高脂饲料喂饲 10天以形成实验性高脂血症大鼠模型 ,同时经口灌胃给予 75、15 0mg·kg-1益脂平胶囊 2 0天 ,测定血液流变学的各项指标。②根据毒理学检测程序[1] ,实验组经口灌胃给予 2 5 0、5 0 0、10 0 0mg·kg-1益脂平胶囊 ,观察其对大鼠的胚胎毒性及致畸作用。结果 :①益脂平胶囊能显著增加实验性高脂血症模型大鼠的红细胞电泳率、显著降低实验性高脂血症模型大鼠的红细胞沉降率 (P <0 .0 5 ) ;能降低实验性高脂血症模型大鼠的全血粘度和血浆粘度 (P <0 .0 5 ) ;②与对照组比较 ,益脂平胶囊实验组孕鼠未见体重增长显著减慢、活产仔数显著减少、死胎率及吸收胎率显著增加以及未见胎仔平均体重、体长、尾长发育指标显著减缓和外观畸形与骨骼畸形明显增加。结论 :①益脂平胶囊可以改善实验性高脂血症大鼠血液流变学指标 ;②在本试验剂量与条件下益脂平胶囊无母体毒性。
Objective:To study the influence of Yizhiping Capsules on hemorrheology of experimental rats with hyperlipoidemia and its embryonic toxicity, teratogenicity. Methods:① The rat experimental hyperlipoidemia models were established by feeding with high lipid diet for 10 days. Meanwhile Yizhiping was taken orally at dosages of 75、150mg·kg -1 , some indices of hemorrheology markers were observed. ②According to the procedure of toxicology, the rat in the experimental groups were administered with Yizhiping capsules by gavage at a dose of 250、500、1000mg·kg -1 respectively, the embryonic toxicity and teratogenicity were observed. Results:① Yizhiping Capsules could significantly raise electrophoresis mobility of red cell of experimental hyperlipoidemia model rats, and lower the rate of red cell deposit and the viscosity of whole blood as well as plasma ( P <0.05); ② Between Yizhiping administered groups and the control group, no significant differences were observed among body weight, body length and tail length, and no increment in incidence of external and skeletal anomalies. Conclusions:① Yizhiping Capsules can improve the abnormal hemorrheology of the experimental hyperlipoidemia model rat; ② Yizhiping Capsules has no maternal toxicity, embryonic and developmental toxicity and teratogenicity under the experimental conditions.
