摘要
目的 :探讨糖尿病慢性溃疡愈合过程中细菌因素的影响以及创面局部使用血小板生长因子 (PDGF)治疗对细菌量变化的影响。方法 :在糖尿病大鼠背部切割圆形创面 ,创面分别涂 PDGF或基质。于伤后不同时间点采样 ,测定创面细菌量及创面面积和伤腔容积。结果 :1创面总细菌量及革兰阴性 (G- )菌量均在伤后第5日达到峰值 ,而后逐渐下降 ,但 G- 菌量下降明显快于总菌量 ,伤后第 7日已降至伤后第 1日水平 ,第 14日已检测不到 ;2伤后早期创面愈合速度较慢 ,第 7日后明显加快 ;这一快速愈合期与 G- 菌菌量快速减少期恰好重叠 ;3PDGF治疗组创面修复速度明显快于基质对照组和单纯致伤组 ,但其创面菌量变化与其余 2组比较无显著差异。结论 :创面 G-菌菌量变化可能影响糖尿病大鼠创面的愈合速度 ;创面使用 PDGF可加快创面愈合速度 。
Objective:To investigate the effect of bacteria on wound healing of diabetes chronic ulcer and influence of treatment with platelet derived growth factor (PDGF) on bacterial counts.Methods:Some circle wounds were made in back of diabetes rats,the wounds were treated with PDGF and matrix.The area size,wound volume and bacterial counts were detected after injury.Results:①The picks of total bacterial counts and the Gram negative (G -) bacterial counts were found on the 5th day after injury and they were decreased gradually afterwards.The decline of G - bacteria counts was faster than that of total bacterial count and it wasn′t detected on the 14th day after injury.②The wound healing was speed up after the 7th day in the wound period, and the quickly healing period of wound was overlap to that of the fast decrease in G - bacteria counts .③The wound healing in PDGF treated group was faster than that in control groups and there was no significant different in wound bacterial counts compared with other two groups.Conclusions:The results suggested that the change of G - bacterial counts in wound might affect the wound healing speed,treatment wound with PDGF would promote the healing,without influence on bacterial count of the wound.
