摘要
目的 :观察不同治疗法则抗癌方对人肝癌细胞株 Hep G2 DNA裂解的影响 ,探讨其抗癌作用机制。方法 :采用分子生物学方法观察益气活血的复方抗纤灵 ( )、行气化瘀的复方 ( )、清热解毒的药物 ( )对人肝癌细胞株 Hep G2 DNA片断、DNA裂解的影响。结果 :Hep G2经中药 、 、 处理 4 8h后 ,电泳分离后 ,低、高剂量 、 方和低剂量的 方均出现明显的 DNA梯状带 ,而 方高剂量和氟脲嘧啶 (5 - Fu)低、高剂量均未见 DNA梯状带 ;Hep G2经中药 、 、 处理 2 4 h后均可诱导 DNA裂解 ,无明显的时间反应 ,而对 、 方有明显的浓度反应。结论 :不同治疗法则的抗癌方诱导人肝癌细胞株 Hep G2 DNA裂解 。
Objective:It was studied that the effect of HepG2 DNA fragmented induced by anti cancer agents of different treatment principle of TCM, having discussed the probable mechanism.Method:it is observed that the DNA fragment of HepG2 cell lines induced by Ⅰ,Ⅱ and Ⅲ anti cancer agents of TCM by moleculer and biologic methods.Results:After HepG2 exposing for Ⅰ,Ⅱ and Ⅲ following 48 bours,electrophoretic analysis of DNA in treated HepG2 showed DNA electrophoresed in a 'Ladder' pattern following Ⅰ(0.5 mg/ml),Ⅱ(0.1 mg/ml,1 mg/ml) and Ⅲ(0.5 mg/ml,0.5 mg/ml).It has previously been show that this pattern of fragments represents multipies of oligonucleosomes.Conclution:That the fragment of HepG2 DNA is induced by anti cancer agents of different treatment principle of TCM is one of the mechanisms of treatment by anti cancer agents of TCM.
