摘要
目的 通过噬菌体展示技术筛选与肝癌细胞特异性结合的抗体。方法 用肝癌细胞系SMMC772 1免疫小鼠 ,提取脾细胞总RNA ,构建单链抗体库 ,从中筛选特异性结合的抗体。结果 构建了一个库容量为 2× 10 5的单链抗体库 ,并筛选到 1个与肝癌细胞系HepG2特异性结合的噬菌体 单链抗体。此单链抗体与HepG2细胞结合滴度比正常肝细胞低 12 5倍以上。结论 本研究成功构建了单链抗体库并筛选到与靶细胞特异性结合的噬菌体 单链抗体。
Objective To select aneibody binding specifically to hepatocyte carcinoma cells by using phage display technique. Methods The recombinant phage antibody system was used to construct a single chain Fv fragment(scFv)cDNA library from total RNA of mice immunized with cell line SMMC7721.Specific antibody was selected from the library.Results A seFv library of 2×10 5 was obtained and a phage scFv named 5~56 which can bind HepG2 cells specifically was found;the titer of scFv 5~56 for binding HepG2 cells was 125 times lower than for binding the normal hepatocyte HL02.Conclusion In this study,a specific phage scFv binding to the target hepatocyte carcinoma cell line was selected from a scFv cDNA library.
