摘要
目的 研究肿瘤来源的热休克蛋白 70 (heatshockprotein 70 ,HSP70 )对荷瘤小鼠的治疗作用 ,为其应用提供参考依据。方法 细胞培养、蛋白质提纯技术、western blot法、毛细管电泳技术和动物实验等。结果 应用 5 μgHSP70能延长小鼠的生存期限 ,平均生存 (2 8.7± 4 .5 )d ,与对照组 (18.5± 3.9)d相比差异显著 (P <0 .0 5 ) ;而 10 μg治疗组的小鼠生存期为 >(6 2 .3± 2 9.1)d ,4 0 %获长期生存 (>90d) ,所接种肿瘤完全消退。与其他组相比 ,差异具显著性 (P <0 .0 1)。结论 肿瘤来源的HSP70具有明确的治疗作用 ,本研究对于研究应用肿瘤来源的热休克蛋白 70治疗人类恶性肿瘤具有较重要的参考意义。
Objective To investigate the therapeutic efficacy of tumor derived heat shock protein 70 to tumor bearing mice, and provide reference for the research of HSP70 administration to treat human cancers. Methods Cell culture, techniques for protein extraction and purification, SDS PAGE, western blot,capillary electrophoresis technique and animal experiment were applied. Results 5 μg HSP70 administration displayed relative anti tumor effect, the survival period of this group was (28.7±4.5) days, it was significant ( P <0.05) as contrasted with that of the control group(18.5±3.9) days. But none of the mice in this group got long period of survival. In 10 μg HSP70 administration group, the survival period of the mice was over (62.3±29.1) days, 40% of the mice survived over 90 days, and the implanted tumors disappeared at the end in the long survived mice, both the survival period and survival rate showed obvious significance as contrasted with those of other groups ( P <0.01). Conclusion Tumor derived HSP70 acted as a potential anti tumor reagent, which offered significant information for HSP70 application to treat human cancers.
