高山红景天多糖RSA的分离纯化和组成分析 被引量:23

摘要 目的 从高山红景天 (RhodiolasachalinensisA .Bor .)根状茎中提出水溶性粗多糖RSP(Rhodiolasachalinensispolysac caride)。从粗多糖中分离出均一多糖并研究其单糖组成。方法 酸性乙醇分级 ,得RS Ⅲ (RhodiolasachalinensispolysaccarideⅢ ) ,脱蛋白 ,脱色 ,DEAE SephadexA 2 5柱层析纯化 ,得一级分RSA(RhodiolasachalinensispolysaccarideA) ,比旋光度法 ,柱层析等方法检验纯度 ,PC和GC测其单糖组成。结果 RSA为均一级分 ,单糖组成为 :Ara ,Rha ,Xyl,Glc ,Gal,GalA ,摩尔比依次为 1.0 0∶3.2 3∶0 .2 6∶0 .34∶0 .84∶10 .2 4。结论 RSA为酸性杂多糖 ,相对分子质量约为 5 0 0 0 0。 OBJECTIVE To obtain a homogeneous polysaccaride RSA from RSP(a water soluble polysaccharide from Rhodila sachalinensis A.Bor.) and study its composition.METHODS RSP was fractionally precipitated with acidic EtOH(pH=2). RS Ⅲ was one of the three fractions.RSA was obtained after RS Ⅲ had been purified by deproteination, decolourization and gel chromatogrophy on DEAE Sephadex A 25. The homogeneity of RSA was proved by gel chromatogrophy on Sepharose CL 4B, gel chromatogrophy on DEAE Sephadex A 25 and specific rotatory power. The composition of RSA was studied by means of GC and PC,etc.RESULTS RSA was homogeneous and consisted of Ara, Rha, Xyl. Glc, Gal, and GalA in the molar ratio of 1.00∶3.23∶0.26∶ 0.34 ∶0.84∶10.24.CONCLUSION RSA is an acidic heteropoly saccharide and its molecular weight is about 50 000.
