百合小孢子发生过程中有关酶活性和组成的变化 被引量:2

CHANGES OF COMPOSITIONS AND ACTIVITIES OF ENZYMES CONCERNED DURING MICROSPOROGENESIS IN LILY

摘要 采用PAG区带电泳方法分析了百合小孢子发生过程中不同阶段细胞内酯酶、过氧化物酶、细胞色素氧化酶、酸性磷酸酶和α—淀粉酶的活性和组成。酯酶、酸性磷酸酶和α—淀粉酶存在于小孢子形成过程中的各个时期。酯酶的活性在细线期到双线期的细胞中较低,终变期以后显著增加、并且有一个新的同工酶带出现,酸性磷酸酶和α—淀粉酶的活性和组成从细线期到第二次分裂末期基本保持稳定,四分体时期分别增加1和2条新的同工酶带。过氧化物酶和细胞色素氧化酶主要出现在减数第一次分裂的前期,中期以后全部消失。SDS—PAG电泳分析揭示双线期以前细胞中的可溶性蛋白一部分在终变期以后消失。终变期以后的细胞内出现许多小分子量的新蛋白质带。关于小孢子发生过程中细胞内生理生化活动本文亦进行了讨论。 The activities and compositions of esterase, peroxidase, cytochromeoxidase, acid phosphatase and α-amylase in the cell of every stage du-ring microsporogenesis have been investigated with PAG electrophoresis.Esterase, acid phosphatase and α-amylase exited in the cell of every sta-ge of microsporogenesis in lily. The esterase was relatively weak in ac-tivity. But after diplotene, the activity of this enzyme greatly increasedand a new isoenzyme band occurred. Both acid phosphatase and α-amylase showed no considerable changes in activity and composition before Tele-phase Ⅱ, but after Telephase Ⅱ they obtained one and two new isoen-zyme bands respectively. The peroxidase and cytochrome oxidase mainlyexited in the cells of Prophase Ⅰ and retained unchangeable during Pro-phase Ⅰ. The SDS-PAG electrophoresis revealed that some of the reso-luble proteins existed in the cells of leptotene-diplotene stages disappe-ared after cells had developed into diakinesis. Some new proteins, themolecular weight of which generally were very small, occurred in thecells of dia kinesis-tetrad stages. The problem about the physiologicaland biochemical activities of cells during microsporogenesis was discussed.
