摘要
采用Southern吸印杂交技术,对人乳头瘤病毒16型(HPV-16)在人宫颈癌活检组织中存在的状况进行了研究。结果提示:①存人宫颈癌组织中存在着HPV-16 DNA序列,并且HPV-16 DNA是整合在肿瘤细胞的基因组中(有的也带有游离的片段)整合的位点是随机的;②不同的人宫颈癌组织HPV-16DNA的含量不同,酶切片段的大小也不同;③HPV-16具有诱导NIH/3T3细胞发生转化的功能,病毒基因亦是整合在细胞染色体DNA中。
HPV-16 DNA homological sequences in biopsy samples from clinical cervical cancer and NIH/3T3 transformed cells are detected by using ^(32)P-dcTp labelled recombinant HPV-16 DNA as a probe with Southern blot Hybridization method. The results imply:in the cervial cancer tissues there are HPV-16 DNA sequences and HPV-16 DNA has integrated into turnour celluler genome(one case was episomal form),whose sites integrate dare random.these varied HPV-16 DNA level and the fragment size of HPV-16 DNA digested by pst Iin different sampls. HPV-16 DNA has a function inducing NIH/3T3 cell to be trausformed,and it also intergrated into the cell chromosome DNA.
