摘要
目的 观察新型乙型肝炎病毒 (HBV)表面抗原中蛋白 (MHBs)核酸疫苗的免疫原性。方法 以新型人体应用载体质粒pSW 3891构建MHBs核酸疫苗 (pSW3891 /MHBs/adr) ,对照组和实验组Balb/c小鼠分别基因枪法免疫对照载体质粒 (pSW 3891 )和MHBs核酸疫苗 ,采用酶联免疫吸附试验检测抗HBs ,LDH释放测定法检测小鼠特异性细胞毒T淋巴细胞 (CTL)杀伤活性。结果 MHBs核酸疫苗可在体外高效表达 ,免疫小鼠后可产生高滴度抗HBs,血清阳性终点滴度达 1∶972 0 0 ,小鼠脾细胞HBs特异性细胞毒性T淋巴细胞 (CTL)杀伤活性达 78 81 %。
Objective To observe immunogenicity of new DNA vaccine encoding for middle hepatitis B virus surface antigen(MHBs).Methods A new DNA vaccine (pSW3891/MHBs/adr) encoding for middle hepatitis B surface antigen was constructed using plasmid pSW3891 which can been used in human.Control and experiment groups of Balb/c mice were immunized with pSW3891 or pSW3891/MHBs/adr by gene gun.Anti HBs in sera of mice were tested by ELISA(enzyme linked immune sorbent assay).Specific cytotoxicity of cytotoxic T lymphocyte(CTL)of mice were detected by LDH release assay.Results pSW3891/MHBs/adr can express in 293T cell line effectively.Mice immunized with pSW3891/MHBs/adr showed strong anti HBs response(ending point>1∶97200) and specific high cytotoxicity of CTL(>78 81%).Conclusion pSW3891/MHBs/adr induced significantly humoral and cellular immune responses in Balb/c mice.
