摘要
对鸡外周血淋巴细胞增殖反应MTT比色法的四个主要因素,即培养时间、ConA浓度、细胞浓度及培养液小牛血清等应用L16(45)四因素四水平正交试验设计,对最佳反应条件进行了研究.结果表明,影响增殖反应的主次顺序为ConA、细胞浓度、培养时间及血清浓度;最佳反应条件为:15μg/ml的ConA、1×107/ml细胞、10%小牛血清及48小时的培养时间.
Four major factors which affect the chicken peripheral lymphocyte proliferate by MTT assay, including the cell culture time and concentration of ConA, lymphocyte and fetal calf serum, has been studied with the method of orthogonal test. The results show that the order of the four factors on the effect of lymphocyte proliferation is the ConA concentration, the cell concentration, the cell culture time and the PCS concentration. The best selection is in the condition of culture for 48h, 15μg/ml ConA, 10%FCS and 1×10~7/ml cell concentration.
