摘要 目的 :观察缺血心肌中环氧化酶 2 (COX 2 )的表达及其抑制剂对前列腺素族代谢产物的影响 ,以了解 COX 2在缺血性心脏病中的作用。方法 :新西兰兔 2 1只 ,随机分为 3组 :正常对照组 (A组 ,n=7) ,心肌缺血组 (B组 ,n=7) ,心肌缺血加罗非昔布组 (C组 ,n=7)。给药 7日后 ,结扎冠状动脉左前降支制作心肌缺血模型。留取血液标本测定血栓素 B2 (TXB2 )和 6酮前列腺素 F1α(6 keto PGF1α) ;取缺血区和右心室非缺血区心肌组织测定 TXB2 和 6 keto PGF1α。对心肌组织行 HE及 COX 2单克隆抗体免疫组织化学染色进行病理观察。结果 :缺血心肌中 COX 2呈高度表达 ;心肌和血清内 TXB2 、6 keto PGF1α水平均较对照组增高 ,而应用罗非昔布干预后 ,则可降低其升高水平。结论 :缺血心肌细胞内 COX 2被诱导表达 ,引起心肌组织中TXB2 和 6 keto PGF1α浓度明显升高 ,COX 2抑制剂可抑制其升高程度。 Objective:To observe the expression of cyclooxygenase2(COX2) in acute ischemia myocardium and the effect of COX2 inhibitor on prostaglandin metabolites in rabbits .Methods:New Zealand rabbits ( n =21) were randomly divided into 3 groups:group A( n =7):normal controls;group B ( n =7),myocardial ischemia,group C( n =7),myocardial ischemia + rofecoxib treatment.After drugs administration for 7 days,a model of acute myocardial ischemia was reproduced by ligation left anterior descending branch for 6 hours in groups B and C.The myocardial section were investigated by HE and monoclonal antibody immunohistochemistry staining,and 6ketoprostaglandin(6ketoPGF 1α ),thromboxane B 2(TXB 2) contents were detected in ischemic and nonischemic tissues.Results:The expression of COX2 was increased in ischemic myocardium.Contents of TXB 2 and 6ketoPGF 1α both in serum and tissues were significantly higher in group B than those in group A.COX2 inhibitor treatment would reduce production of prostaglandin metabolites in ischemic myocardium.Conclusions:Acute myocardial ischemia may result in the elevation of TXB 2 and 6ketoPGF 1α contents,which can be inhibited by COX2 inhibitor in serum and ischemic myocardium.
